Effects of Polychlorinated Biphenyl (PCB)

Effects of Polychlorinated Biphenyl (PCB) Polychlorinated Biphenyl (PCB) is a persistent organochlorine that has 209 congeners and can be divided into two categories, dioxin-like and non-dioxin-like. Although these two categories have different structures and toxicological profiles, both have an adverse impact on the human health [1]. Since PCB is a stable chemical and can withstand extreme temperature and pressure, it was heavily used back in 1929 for electrical equipment [2]. While the production of PCB was ceased in 1977, traces of PCB can still be observed on the Great Lakes for it was once used as a disposal site along with the possibility of accidental leaks. From lakes, it can move into different waterways and later be passed on by organisms. Long-range-transport of PCB vapours through air currents also functions as a dispersion mechanism in the ambient air [3] [4] [5]. Anyone can be susceptible depending on the rate of the exposure and the amount of stressor a person had been exposed to. PCB can enter the system thro ugh ingestion, inhalation and dermal contact, all of which can essentially bioaccumulate and inhibit the bodys natural processes in the event that the receptor had exceeded the threshold. PCB has been linked to endocrine dysfunction, reproduction deficiencies, chloracne, liver damage and kidney cancer [6]. Recognizing credible sources is a key factor in toxicity assessment. This allows the reader to acquire more information about the chemical of concern and make an informed decision. Some credible sources are listed below: Agency for Toxic Substances and Disease Registry European Chemicals Agency Health Canada United States Environmental Protection Agency World Health Organization     Ã‚   3.1. What happens when PCB enters the body? Humans absorb PCB through inhalation, ingestion and dermal contact. On bioassays conducted, it was observed that animals absorbed PCB well when administered orally and less absorbed when exposed through dermal contact. While inhalation is an accounted pathway for PCB, there is inadequate data to identify the absorption rates. Upon absorption, PCB is dispersed into the lipophilic cell membranes in the gastrointestinal tract. It then circulates into the blood vessels and enters the lymphatic system. Due to the lipophilic nature of PCB molecules, it is readily absorbed by the fatty tissue. In autopsies conducted, the liver, adipose tissue, brain and skin usually contains higher concentrations of PCB. Elimination of PCB can be through excretion of feces, urine and breast milk [7]. When exposed to PCB, symptoms include severe acne, swelling of the upper eyelids, discolouring of the nails and skin, numbness in arms and/or legs, weakness, muscle spasms, chronic bronchitis, and neurological complications [6]. 3.2. Acute and Chronic Effects PCB exhibits both acute and chronic effects. Although it demonstrates low potential in producing acute effects, however, due to the risk it poses in human health in the long-term, it is still deemed to be a public health concern. Skin lesions particularly chloracne has been observed during occupational exposures. Nausea and headaches were also reported yet this could be primarily due to the solvents in the PCB mixtures. Subsequently, long-term exposure to PCB has been linked to reproductive deficiencies, liver damage and kidney cancer [8]. 3.3. Threshold vs Non-threshold PCB has shown evident carcinogenicity in animals and therefore is considered to be non-threshold substance by USEPA. The data for human carcinogenicity for PCB mixture is deemed to be inadequate but suggestive. Essentially, PCB can also be classified as threshold substance because of its non-cancer potential in lower concentrations and short-term exposure [9].   3.4. What effects does it have? Carcinogenic US EPA labeled PCB as Group B2, a probable human carcinogen. This was based on the experiment of exposure of rats to several mixtures of PCB that are often found in the environment [10]. On a study published in 1976, occupational exposure to PCB increased the mortality from cancer. However, the outcomes were mixed since the study only found limited deaths and incomplete exposure assessments [7]. Mutagenicity There were insufficient data involving the mutagenicity of PCB [7]. A bacterial test which distinguishes chemical mutation effects was conducted on PCB. Although PCB demonstrated negative results, this was not uncommon since PCB has complex compounds and pathways [11]. Reproductive Toxicity Oral administration of PCB on animals has displayed toxicity effects but dermal and inhalation exposure were not available. Exposure of rats and mice to high doses of PCB had produced several effects. The effects documented were decreased conception rate, pro-longed menstruation, decreased litter size and reduced sperm count. Rats and mice that were exposed to PCB while lactation suggested long-lasting adverse effect on the offspring. On another study, PCB is also associated to estrogen-related effects [7]. 4.1. Bioassay Overview The slope factor of PCB was based upon the studies conducted by Brunner et al. (1996) and Norback and Weltman (1985). A group of male and female Sprague-Dawley rats were exposed to different Aroclor. Groups of 50 female or male rats were orally administered with 25, 50 or 100 ppm Aroclor 1260 or 1254; 50 or 100 ppm Arcolor 1242; or 50, 100, or 200 ppm Aroclor 1016. At 104 weeks, the rats were killed. Liver adenomas or carcinomas were discovered in all female rats for all Aroclors and male rats for Aroclor 1260. While thyroid gland follicular cell carcinoma was evident on male rats, female rats also exhibited mammary tumors at a later age. On another study, male and female Sprague-Dawley rats were exposed to 0 or 100ppm of Aroclor 1260 for 16 months and subsequently reduced to 50 ppm for another 8 months. Tests were executed in different months to understand the changes in the structure of the rats. The intensity of the damage on the liver increases as you progress through the months. On the 29th month, most of the female rats displayed carcinoma and neoplastic nodules whereas male rats had lesser incidence [10] . 4.2. Published Benchmark A tiered approach was used by USEPA to determine the cancer potency of PCB. Upper-bounds provide precision while central-estimates are appropriate for estimating risks. Potency values are dependent on the exposure pathway. For exposure pathways with increased risks, we use High Risk and Persistence. Exposure pathways that decrease risk are appropriate on Low Risk and Persistence. Lowest Risk and Persistence is developed from Aroclor 2016, therefore it is appropriate for congeners with more than for chlorine. The slope factor can be generated by dividing 0.10 by ED10 (Estimated dose associated with 10% increased incidence, in mg/kg-d) [12]. High Risk and Persistence Low Risk and Persistence Lowest Risk and Persistence Upper-Bound Slope Factor 2.0 per (mg/kg)/day 0.4 per (mg/kg)/day 0.07 per (mg/kg)/day Central-Estimate Slope Factor 1.0 per (mg/kg)/day 0.3 per (mg/kg)/day 0.04 per (mg/kg)/day Criteria for use Food chain exposure Sediment or soil ingestion Dust or aerosol inhalation Dermal exposure, if an absorption factor has been applied Presence of dioxin-like, tumor-promoting, or persistent congeners Early-life exposure (all pathways and mixtures) Ingestion of water-soluble congeners Inhalation of evaporated congeners Dermal exposure, if no absorption factor has been applied Congener or isomer analyses verify that congeners with more than 4 chlorines comprise less than 1/2% of total PCBs. Figure 1 Tiers of Human Slope Factors For Environmental PCB [13]. Uncertainty Factors It is important to know that not everyone responds equally to a given chemical. Populations with higher sensitivity may be associated with increased risks. Also, the genetics and lifestyle of a person must be accounted as this pose potential change on the dose-response trend. Furthermore, it is essential to recognize that the mixtures of PCB tested in animals are Aroclor mixtures which have been pre-determined and not environmental mixtures that had bioaccumulated which are generally more potent [10]. Figure 1 shows that the slope factor is dependent upon the exposure pathway. Although these are just estimates, scientists have dedicated their time to conduct bioassays where we can extrapolate the data to potentially understand the dose that can harm humans. However, it is also crucial to know that environmental processes can alter the PCB concentrations, therefore it is best to be educated on how you can be exposed to PCB and how you can avoid this. To improve the assessment, studies concerning sensitive population should be further considered and assessed. Since PCB concentrations are altered in the environment, animals or humans that are already exposed could be used to investigate and compare the differences with the experiments done in the laboratory. In addition, it was not mentioned why rats were chosen as experiment parameters, analyzing parameters used could help readers discern why rats are relevant for the study. [1] P. Heikkinen, . L. T. Microorganisms Relevant To Bioremediation

Microorganisms Relevant To Bioremediation Introduction Bioremediation is a technology that utilizes the metabolic potential of microorganisms to clean up contaminated environments. One important characteristic of bioremediation is that it is carried out in non-sterile open environments that contain a variety of organisms. Of these, bacteria, such as those capable of degrading pollutants, usually have central roles in bioremediation, whereas other organisms (e.g. fungi and grazing protozoa) also affect the process. A deeper understanding of the microbial ecology of contaminated sites is therefore necessary to further improve bioremediation processes. In the past two decades, molecular tools, exemplified by rRNA approaches, have been introduced into microbial ecology; these tools have facilitated the analysis of natural microbial populations without cultivation. Microbiologists have now realized that natural microbial populations are much more diverse than those expected from the catalog of isolated microorganisms. This is also the case for pollutant-degrading microorganisms, implying that the natural environment harbors a wide range of unidentified pollutant-degrading microorganisms that have crucial roles in bioremediation. This article summarizes the results of recent studies of microbial populations that are relevant to bioremediation. Molecular ecological information is thought to be useful for the development of strategies to improve bioremediation and for evaluating its consequences (including risk assessment). Molecular tools are especially useful in bioaugmentation, in which exogenous microorganisms that are introduced to accelerate pollutant biodegradation need to be monitored. This article discusses recent examples of the successful application of molecular ecological tools to the study of bioremediation. Microorganisms relevant to methane oxidation Traditionally, studies on pollutant biodegradation have been initiated by the isolation of one or more microorganisms capable of degrading target pollutants; however, conventional isolation methods have resulted in the isolation of only a fraction of the diverse pollutant-degrading microorganisms in the environment. In addition, most isolated organisms have shown pollutant-degradation kinetics that differ from those observed in the environment. For example, laboratory-cultivated methanotrophs exhibit half-saturation constants for methane oxidation which are one to three orders of magnitude higher than those observed in soil. Using molecular phylogenetic analyses of isotope-labeled DNA, (Radajewski et al.) successfully identified two novel methanotrophs that actively degrade methane under environmental conditions. Molecular approaches that target the 16S rRNA gene (16S rDNA) and genes encoding enzymes involved in key metabolic steps (e.g. those encoding particulate methane monooxygena se) have been applied to the analysis of methanotrophs in rice field soil, lake sediments and forest soil. Methanotrophs are considered to be important for reducing the emission of methane, a greenhouse gas, from soil and sediment. In addition, methanotrophs co-metabolize trichloroethylene (TCE); therefore, TCE bioremediation often employs methane injection as a means to stimulate the TCE-degrading activity of indigenous methanotrophs (i.e. methane biostimulation). Methanotrophs which occurred at a methane biostimulation site were recently analyzed using denaturing gradient gel electrophoresis (DGGE) of polymerase chain reaction (PCR)-amplified 16S rDNA and soluble methane monooxygenase gene fragments. Marine petroleum hydrocarbon degradation Molecular ecological approaches have also been used to analyze bacterial populations that occur in petroleum-contaminated marine environments. Spilled-oil bioremediation experiments conducted at a sandy beach found that phylotypes affiliated with the subclass of Proteobacteria appeared in the DGGE fingerprints obtained for oiled plots but not in those for unoiled plots, suggesting their importance in spilled-oil bioremediation. Another oil-spill experiment conducted at a beach in the Norwegian Arctic showed that 16S rDNA types affiliated with the ÃŽÂ ³-Proteobacteria, especially those belonging to the Pseudomonas and Cycloclasticus groups, were abundant in fertilized oil sands. Microbial populations which occurred in seawater after supplementation with petroleum and inorganic fertilizers have been analyzed using rRNA approaches; it was reported that bacterial populations belonging to the Proteobacteria and the genus Alcanivorax showed accelerated growth. These studies have indicat ed that some groups of bacteria commonly occur in oil-contaminated marine environments, although other populations change under different environmental conditions. Anaerobic petroleum hydrocarbon degradation As petroleum hydrocarbons are persistent under anaerobic conditions, their contamination of groundwater is a serious environmental problem. The microbial diversity in a hydrocarbon- and chlorinated-solvent contaminated aquifer undergoing intrinsic bioremediation was assessed by cloning and sequencing bacterial and archaeal 16S rDNA fragments. This study detected phylotypes that were closely related to Syntrophus spp. (anaerobic oxidizers of organic acids with the production of acetate and hydrogen) and Methanosaeta spp. (aceticlastic methanogens), suggesting their syntrophic association. Phylotypes affiliated with candidate divisions (that do not contain any isolated organisms) were also obtained in abundance from the contaminated aquifer, although their physiology is completely unknown. A similar syntrophic association of bacteria and archaea has also been reported in a methanogenic enrichment that slowly degrades hexadecane. Likewise, a toluene-degrading methanogenic consortium was characterized by rRNA approaches. The consortium comprised two archaeal species related to the genera Methanosaeta and Methanospirillum, and two bacterial species, one related to the genus Desulfotomaculum and the other unrelated to any previously described genus. Fluorescence in situ hybridization (FISH) with group-specific rRNA probes was used to analyze a denitrifying microbial community degrading alkylbenzenes and n-alkanes; the Azoarcus/Thauera group was found to be the major bacterial group. Bacteria affiliated with the ÃŽÂ µ-Proteobacteria were found to grow in petroleum-contaminated groundwater which accumulated at the bottom of underground crude-oil storage cavities. Microbial communities associated with anaerobic benzene degradation under Fe(III)-reducing conditions in a petroleumcontaminated subsurface aquifer were also analyzed by DGGE analysis, and it has been suggested that Fe(III)- reducing Geobacter spp. have an important role in the anaerobic oxidation of benzen e. The available electron acceptors are the principal determinants for the types of microorganisms that occur in anaerobic environments, and microbial populations identified in the above papers are considered important for petroleum hydrocarbon degradation in subsurface environments under the respective conditions. On the basis of these results, future developments in anaerobic hydrocarbon bioremediation are anticipated. It is noteworthy that phylotypes that are only distantly related to known genera are often detected as major members of the anaerobic communities, suggesting that parts of anaerobic hydrocarbon biodegradation processes remain unidentified. Polycyclic aromatic hydrocarbon degradation Polycyclic aromatic hydrocarbons (PAHs) are compounds of intense public concern owing to their persistence in the environment and potentially deleterious effects on human health. A soil-derived microbial consortium capable of rapidly mineralizing benzo[a]pyrene was analyzed by DGGE profiling of PCR-amplified 16S rDNA fragments The analysis detected 16S rDNA sequence types that represented organisms closely related to known high molecular weight PAH-degrading bacteria (e.g. Burkholderias, Sphingomonas and Mycobacterium),although the degradation mechanisms have yet to be resolved. In soil environments, the reduced bioavailability of PAHs due to sorption to natural organic matter is an important factor controlling their biodegradation. Friedrich et al. reported that different phenanthrene-degrading bacteria occurred in soil enrichments when different sorptive matrices were present. It has also been shown that the application of surfactants to soil enrichments that degrade phenanthrene a nd hexadecane altered the microbial populations responsible for the degradation. These results have common implications for bioremediation; that is, nature harbors diverse microbial populations capable of pollutant degradation from which a few pollutant-degrading populations are selected according to bioremediation strategies. Metal bioremediation Because of its toxicity, metal contamination of the environment is also a serious problem. Recent studies have applied molecular tools to the analysis of bacterial and archaeal populations that are capable of surviving in metal-contaminated environments. Bacterial communities in soil amended for many years with sewage sludge that contained heavy metals were assessed using rRNA approaches, including FISH and cloning and sequencing. The study found that two sequence groups affiliated with the Proteobacteria and Actinobacteria were frequently obtained from clone libraries from the metal-contaminated soil, although most Actinobacteria sequences showed low similarity (

Corporations and How They Govern Our Lives Essay -- Business, Marketin

The existence of large corporations has become a way of life for most of us. We have grown up around these companies as they have come to dominate our lives. These large corporations have taken over many facets of our lives. The purchasing power of large corporations has given them an unprecedented degree of power over many aspects of our lives. â€Å"They determine what we eat, what we watch, what we wear, where we work and what we do.†(Bakan,5). Corporations, who are not considered humans, are raising our children and dictating what is eaten, bought, worn and what is cared about. In times gone by, humans were shaped by their family and culture but now we are manipulated and shaped by zealous marketers whose definitive goal is to maximize profits with a total disregard to any social responsibility. There is, however, a growing trend of corporations who engage in community work and are enthusiastic on protecting the environment. â€Å"IKEA focuses heavily on sustaina ble forestry as you would expect from a furniture supplier that uses a lot of wood and is changing the lives of 80,000 children which is still a big accomplishment.† (Barner, 59-60). Ultimately, the employees of such corporations as well as the consumer of these companies want to actively participate in the social responsibility initiatives of the corporation they work and buy from. Corporations, through the use of their excessive marketing strategies and with relatively little risk placed upon them, corporations are able to project their power and influence on society as a whole as well as upon all aspects of governments. Since the early twentieth century, large corporations have become part of our economic makeup. A corporation is a separate legal entity that... ...emains, can a corporation work for both the betterment of society and at the same time make a profit? There are a handful of corporations that are coming around and understanding that if they want to be around for the long term, they need to make adjustments to the way they operate their businesses. With all the recalls in the media, Consumers are getting fed up with these establishments because these recalls were primarily caused by shortcuts on the company’s part and total disregard for our health and welfare. Today, mindfulness and genuine concern for the consumer’s health and safety needs to be a Corporation’s number one priority mission if they are to continue its profitability. Ultimately, at the end of the day, the thing that most of us really want is a decent place to live, a secure income, untainted food, proper education and good health care.

Malcolm X :: essays research papers

Malcolm's life is a Horatio Alger story with a twist. His is not a "rags to riches" tale, but a powerful narrative of self-transformation from petty hustler to internationally known political leader. Born in Omaha, Nebraska, the son of Louise and Earl Little, who was a Baptist preacher active in Marcus Garvey's Universal Negro Improvement Association, Malcolm, along with his siblings, experienced dramatic confrontations with racism from childhood. Hooded Klansmen burned their home in Lansing, Michigan; Earl Little was killed under mysterious circumstances; welfare agencies split up the children and eventually committed Louise Little to a state mental institution; and Malcolm was forced to live in a detention home run by a racist white couple. By the eighth grade he left school, moved to Boston, Massachussetts, to live with his half-sister Ella, and discovered the underground world of African American hipsters. Malcolm's entry into the masculine culture of the zoot suit, the "conked" (straightened) hair, and the lindy hop coincided with the outbreak of World War II, rising black militancy (symbolized in part by A. Philip Randolph's threatened March on Washington for racial and economic justice), and outbreaks of race riots in Detroit, Michigan, and other cities (see Detroit Riot of 1943). Malcolm and his partners did not seem very "political" at the time, but they dodged the draft so as not to lose their lives over a "white man's war," and they avoided wage work whenever possible. His search for leisure and pleasure took him to Harlem, New York, where his primary source of income derived from petty hustling, drug dealing, pimping, gambling, and viciously exploiting women. In 1946 his luck ran out; he was arrested for burglary and sentenced to ten years in prison Malcolm's downward descent took a U-turn in prison when he began studying the teachings of the Lost-Found Nation of Islam (NOI), the black Muslim group founded by Wallace D. Fard and led by Elijah Muhammad (Elijah Poole). Submitting to the discipline and guidance of the NOI, he became a voracious reader of the Qu'ran (Koran) and the Bible. He also immersed himself in works of literature and history at the prison library. Behind prison walls he quickly emerged as a powerful orator and brilliant rhetorician. He led the famous prison debating team that beat the Massachusetts Institute of Technology, arguing against capital punishment by pointing out that English pickpockets often did their best work at public hangings!

Nursery Rhymes Essay

So here I am, really scared and nervous, but here’s a secret my mum told me, adults are just as anxious talking in public, the trick is to picture everyone in clown suits; my Apa’s red nose really suits her. Hmm, my speech is supposed to be humorous and as a 6 year old nothing is more hilarious as old nursery rhymes I mean can you really picture a huge egg perched on a wall, and why on earth did the king send his army to help humpty when maybe a chef would’ve done instead. Again, why on earth was the king involved? Then you get insy wincy, for being a spider he wasn’t a very bright spark. He firstly climbed up a water spout.. let’s see, cloudy day, water sprout..rain..light bulb..ting†¦but no, he goes and climbs up the spout. Again next time use the wall, he must have been a male, duh.. We pass the old age home every day on our way to school. I always feel sorry for these poor people; I mean how can people leave their mom’s there. But then I think about the old women who lived in a shoe.. Now that’s really sad, sure do hope it’s a really big shoe. Nursery rhymes are really silly, but if you can’t beat them join them. There’s my version: There was an old woman who lived in a shoe. The place was disgusting and smelled like pee-ewe! The windows were drafty. The roof was a leaker. But that’s what you get When you live in a sneaker. Humpty Dumpty sat in a tree. Humpty Dumpty got stung by a bee. He fell out and hit his head, and now he thinks his name is â€Å"Fred.† The tighty-whity spider went down the waterslide. Got a water wedgie halfway down the ride. Jumped up and screamed and ran around in pain. Now the tighty-whity spider will not do that again.

Language and Arthur Millers skills as a dramatist Essay

â€Å"A view from the bridge† is a two-act tragic play by Arthur Miller in the mid 50s. This play puts light on the working American-Sicilian population, in particular the Carbone family, the head of which is Eddie Carbone. In this essay I will be analysing Eddie Carbone’s character whilst occasionally making comments on the language and Arthur Miller’s skills as a dramatist. Eddie is a simple, straight forward man that lives with his beloved wife and niece. Alfieri describes Eddie as a â€Å"long shore man†. He is very protective over his orphaned niece, Catherine. Arthur Miller uses informal language to show Eddie’s social class. It is worthy to note that Arthur Miller was skilfully able to come down to the status of long shore man and was able to produce speech successfully in their tone and language. Eddie is portrayed as generous, but at the same time selfish and self-protective. This is since, although he is generous to offer his home to two of his wife’s cousins, Marco and Rodolfo, he believes he is Superior. This is shown as he reminds Beatrice not to offer them his bed and he is very jealous of anyone that gets too close to Catherine. This jealousy also adds to his hardship in accepting Rodolfo and Catherine’s relationship, another reason to this envy is his over-protective nature. He has brought up Catherine as his own daughter and feels afraid to let her grow up, so he cannot face her having any relation to other men. Eddie’s wife, Beatrice’s is unhappy with Eddie’s attitude towards Catherine, which soon starts to create a rift between the couple. â€Å"It’s been many days and nights since you haven’t slept with me†; shows that Eddie has been so caught up in Catherine and making sure she does not fall for Rodolpho, that he begins to forget about his duties as a husband. Further into the play, Eddie begins to feel another emotion towards Catherine: sexual attraction. This is shown by many things which happen throughout the play. He quickly begins to feel extremely jealous of the immediate impression Rodolfo makes on Catherine. The stage directions state at a point, â€Å"He looks at [Catherine] like a lost boy†, when she reveals her feeling for Rodolfo to him, but he is unable to admit it to everybody else. Furthermore, Eddie becomes so obsessed with Catherine, that he has no control over his emotions; for example Catherine begins to like Rodolpho, Eddie cannot control himself and has to find a way to vent his anger, so He decides to hurt Rodolpho, by using an exercise of teaching him to box,:†you ever do any boxing†? Eddie, at this point, feels so remorseless, that even when Rodolpho refuses to hit him, â€Å"I don’t want to hit you Eddie,† he stills feels like the need to hit him, showing how obsessed he is. Moreover, Eddie becomes too interested into the relationship between Catherine and Rodolpho, that he involves the Immigration Bureau, this is because he could not spilt the true love apart. In the eyes of the audience and the other characters, it causes Eddie’s remaining honours to be thrown in the dirt. As in any community, even though there may be accused, the government should not be called in, otherwise the one, who spread the word, shall not be respected by all others and therefore, he becomes an out-cast. His redemption occurs towards the end of the play. His honour is restored with his own death. This is as he fights with Marco and ultimately dies, but he accepts his fate, and therefore restoring his respect and honour. In this part of the play, he also shows how he had planned the fight and was so obsessed that he brought a knife and fought like a coward. This brought his redeemed respect again to an all time low. Society respects a man, who acts like a man, and Eddie in this part, fights like a coward, which is unacceptable in the society. In conclusion, Eddie, although being a real man at the start, ends his life in a shameful and cowardly way. In my analysis, I have shown how Arthur Miller portrays this in an effective and skilful way, which shows his brilliant ability as a writer. I feel that ‘A View from the Bridge’ is a very high-quality book with a interesting and deep story, giving it full marks in my view.
